Description
- Recognizes human NGAL
- NGAL (Neutrophil gelatinase-associated lipocalin, Lipocalin-2, LCN2) is a secreted glycoprotein that belongs to lipocalin protein family
- NGAL exists as a monomer, homodimer, disulfide-linked heterodimer in cells, covalently conjugated with gelatinase in order to decrease degradation
- Roles include iron trafficking, chemotactic, promote cell differentiation and proliferation
- Found in human neutrophils and expressed in tissues containing renal, uterus, liver and spleen, cardiomyocytes, neurons, immune cells
- NGAL is a biomarker of cardiovascular diseases, ischemic stroke, acute renal injury, inflammation, metabolic diseases
- Detected in human serum, saliva, urine
Process:
- Antibody specific for NGAL pre-coated onto microwells
- NGAL protein in samples captured by the coated antibody after incubation
- After extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for NGAL is added to detect captured NGAL protein
- For signal development, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
