  • Upregulated in wide variety of solid tumors (melanoma, ovarian, lung, glioblastoma, breast, pancreatic cancers)

Process:

• Antibody specific for PD-L1 pre-coated onto microwells
• PD-L1 protein in samples captured by the coated antibody after incubation
• After extensive washing, another antibody specific for PD-L1 is added to detect captured PD-L1 protein
• For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
