Description

The Mouse Tissue Inhibitor of Matrix MetalloProteins 1-Cell Lysate (Ms TIMP-1) ELISA quantitates Ms TIMP-1 in mouse cell or tissue lysates. The assay will exclusively recognize both natural and recombinant Ms TIMP-1. Principle of the method The Mouse TIMP-1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Tissue inhibitor of metalloproteinases-1 (TIMP-1) and Tissue inhibitor of metalloproteinases-2 (TIMP-2) have similar properties, specifically in inhibiting enzymes of matrix metalloproteinase family, and are thought to be of great importance in the maintenance of connective tissue integrity. TIMP-1 forms a complex of 1:1 stoichiometry with activated interstitial collagenases, activated stromelysin, active form of 72kDa Type IV collagenase (also known as MMP-2 or gelatinase A), and latent and active forms 92kDa Type IV collagenase (also known as MMP-0 or gelatinase B). TIMPs inhibit the proteolytic invasiveness of tumor cells and normal placental trophoblast cells.
