pYSG-IGA yeast expression vectors
The pYSG-IBA3 vector allows the high-level expression of proteins in yeast. The vector carries the copper inducible promoter (CUP1) for controlled high-level expression. The expressed recombinant protein will be localized in the cytoplasm.
- High-level expression in yeast with high copy number
- Favors correct protein folding and the production of soluble proteins - inclusion bodies rarely form
- Tightly regulated expression due to Cu++-inducible CUP1 promoter
- Ampicillin resistance gene and markers Leu2-d and URA3 in every vector
- Leu2-d auxotrophy marker for selection to increase plasmid copy number during expression
- URA3 auxotrophy marker for selection after transformation
StarGate™ System
IBA developed StarGate to provide a simple, fast and highly efficient cloning process for genes of interest (GOI) for use in functional expression, purification, detection, immobilization or separation of recombinant proteins:
- Minimal extra modification of the gene due to short combinatorial sites
- Inherent high-level cloning efficiency due to a directed reaction (no equilibrium)
- GOIs are cloned to obtain a Donor Vector; 2 or more genes can be combined using Fusion Cloning
- Genes are then transferred into Acceptor Vectors that provide optimal genetic surroundings
- The final expression vector, the Destination Vector, is then placed into the respective host
