The CBQCA Plus Protein Quantitation Kit is a specialized tool designed for the accurate quantitation of proteins in solution. This kit is a refined version of the CBQCA Protein Quantitation Kit (Cat. No. C6667), with a significant modification: potassium cyanide, a toxic compound, has been replaced with mandelonitrile, a less-toxic alternative. This change does not compromise the kit's performance, maintaining the same level of sensitivity and accuracy as its predecessor.

The CBQCA Plus Protein Quantitation Kit exhibits high sensitivity and is capable of detecting as little as 50 ng of bovine serum albumin (BSA) in an assay volume of 100–200 μL. This sensitivity allows for precise measurements even in low-protein samples. Moreover, the kit is compatible with detergents, a feature that enhances its versatility. This compatibility makes it suitable for accurate quantitation of proteins in the presence of lipids, membrane fractions, or detergents, as well as for lipoproteins and small peptides.

The kit includes all necessary components for performing 300–800 assays, depending on the concentration of substrate used. The resulting fluorescent product is maximally excited at 465 nm and has an emission maxima at 550 nm. It is designed for use with a fluorescence microplate reader, but can also be adapted for use with a standard fluorometer or minifluorometer after dilution of the reaction mixture.

There are two main differences between the two kits. Firstly, the CBQCA Protein Quantitation Kit (Cat. No. C6667) contains a potassium cyanide (KCN) component (component C). This component has been removed in the CBQCA Plus Protein Quantitation Kit (Cat. No. A66522) and replaced with mandelonitrile which serves as a less toxic alternative to KCN without compromising assay performance. Secondly, the CBQCA Plus Protein Quantitation Kit includes one additional component compared to the CBQCA Protein Quantitation Kit, a BupH Borate Buffer Pack.

What is the shelf life of the BupH Borate Buffer Pack after reconstitution?

When properly reconstituted using a clean water source and good handling technique, the BupH Borate Buffer is stable for approximately 3-6 months when stored at 4 degrees C. For longer-term stability, we recommend adding sodium azide to a final concentration of 0.05% (w/v).

What is the working range for the CBQCA Plus Protein Quantitation Kit?

Yes, the product is still good to use. We ship many of our products at a higher temperature than the storage recommendation. The product will be stable for at least one week at the shipping temperature when in the original, unopened packaging. For long-term stability, the product should be placed at the recommended storage temperature upon receipt.

Can I use the CBQCA Plus Protein Quantitation Kit to quantitate small peptides?

We do not recommend using the CBQCA assay for quantitating small peptides since the BSA standard used is not reflective of a peptide sample. A better assay to use would be the Pierce Quantitative Fluorometric Peptide Assay (Cat. No. 23290) or the Perce Quantitative Colorimetric Peptide Assay (Cat. No. 23275).

Where can I find the expiration date for the CBQCA Plus Protein Quantitation Kit?

The CBQCA Plus Protein Quantitation Kit does not have an expiration date. The kit is guaranteed for 12 months from date of shipment, when stored as indicated in the user manual. Once the CBQCA is reconstituted in DMSO, the working reagent has a shelf life of 6 months.

Should the CBQCA Plus Protein Quantitation Kit be read from the top or bottom on a plate reader?

The CBQCA assay is a suspension assay. Suspension assays should always be read with a top read on a plate reader for best sensitivity. Bottom reads will give reduced sensitivity.

How do I know if the contaminants in my samples are compatible with the CBQCA Plus Protein Quantitation Kit?

For substances that do not contain primary amines and that we have not validated to be compatible with the CBQCA assay, we recommend preparing two standard curves, one with the substance spiked in at the desired concentration in the assay and one without the substance spiked in. If the substance does not significantly affect the quantitation of the assay, then you can use it in the assay. You should prepare the standard curve with the substance spiked in at the same concentration as used with the samples, so that the standards are under the same conditions.

What is the compatibility of the CBQCA Plus Protein Quantitation Kit with urea or guanidine?

Both urea and guanidine contain primary amines, so neither are compatible with the CBQCA assay.

Can the CBQCA Plus Protein Quantitation Kit be used on the Qubit Fluorometer?

The excitation/emission (Ex/Em) wavelengths of CBQCA are 465 nm and 550 nm respectively, which would fit within the blue excitation range of 430-495 nm and green emission range of 510-580 on the Qubit instruments, so it should be possible to set up a MyQubit assay for this reagent.
