General Description
- 3,3′-Diaminobenzidine is a derivative of benzene and the precursor to polybenzimidazole
Applications
- DAB is mainly used to visualize peroxidase activity. During this chemical reaction, DAB is a hydrogen donor in the presence of peroxide. When DAB is oxidized, an insoluble brown end-product is produced.
- DAB has been used for staining and oxidative detection of mitochondria
- It has also been utilized for electron microscopy applications

Frequently Asked Questions (FAQs)
1. Remove the bottle from the refrigerator and allow to stand at room temperature (~20 degrees C) for 15-30 min.
2. Dissolve five tablets containing 10 mg DAB in 10 mL of distilled water at room temperature. Do not use metal forceps during handling of the tablets, since metal ions catalyze the oxidation process. Protect the solution from direct sunlight.
3. Some fine particulate matter will remain in the suspension even after the solution is thoroughly mixed. This will not affect your results, however if preferred, the solution can be filtered using Whatman No. 2 paper.
4. Add 4 urea peroxide tablets (Acros Cat. No. 32807) to 10 mL of the substrate solution. Alternatively add 40 µL 30% hydrogen peroxide (H2O2) and mix. This solution should be used within 2 hours.
