Description
- Kit provides everything needed to covalently label proteins with a dibenzylcyclooctyne (DBCO) group for copper-free click chemistry applications
- Contains all necessary materials to perform five labeling reactions and subsequent purification for removal of excess DBCO reagent
- TFP-PEG4-DBCO is a heterobifunctional crosslinker which contains an amine reactive tetrafluorophenyl (TFP), and an azide reactive dibenzylcyclooctyne (DBCO) with a water solubilizing PEG4 spacer in between
- It readily reacts with the primary amines found on proteins to form a covalently attached DBCO group, which can be further reacted with an azide labeled coupling partner (supplied separately) to create diverse bioconjugates
- In the first conjugation step, the TFP group reacts with a primary amine on a protein to form a stable amide bond and a covalently bound DBCO moiety
- Subsequent purification removes excess crosslinker from solution which may interfere with the second labeling step
- Once a protein is DBCO labeled and purified, it can undergo a copper free strain-promoted azide-alkyne cycloaddition (SPAAC)
- Label proteins (20 to 150 kDa) with concentrations ranging from 0.5 to 5 mL
- TFP esters are more hydrolytically stable than NHS esters enabling less reagent to be used
- Labeling and purification can be completed in as little as 90 minutes with 30 min hands-on time
