Hersintuzumab is a novel humanized anti-HER2 monoclonal antibody designed to target HER2-overexpressing cancers. The ERBB2 gene, located on chromosome 17 at position 17q12, encodes the ErbB2 protein, also known as HER2 (human epidermal growth factor receptor 2). The ErbB2 protein is a member of the receptor tyrosine kinase family and is characterized by its extracellular domain, transmembrane segment, and intracellular tyrosine kinase domain. Structurally, the extracellular domain of ErbB2 forms a homodimer through a back to head interaction, which is critical for its function. ERBB2 is best known for its role in the pathogenesis of various cancers, particularly breast cancer. Overexpression or amplification of the ERBB2 gene leads to excessive ErbB2 protein on the cell surface, resulting in constant cell growth signaling. Unlike other members of the ErbB family, no specific ligand has been found for ErbB2, and its activation often involves heterodimerization with other ErbB family receptors such as EGFR. The activation of these receptors triggers downstream signaling pathways that promote cell proliferation and survival.
