Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous CRP

  • Recognizes human CRP
  • C-reactive protein (CRP) is an acute-phase serum protein predominantely synthesizes by liver in response to interleukin-6 during inflammation
    • Ability to react with C polysaccharide of Streptococcus pneumoniae
    • Annular, pentameric protein belongs to pentraxin family of proteins
    • Displays functions associated with host defense: promotes agglutination, bacterial capsular swelling, phagocytosis and complement fixation through calcium-dependent binding to phosphorylcholine
    • Used as inflammation marker

Process:

  • Antibody specific for CRP pre-coated onto microwells
  • CRP protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for CRP is added to detect captured CRP protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
