Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous CYCS

  • Recognizes human CYCS
  • CYCS (Cytochrome c) is a 12-15 kDa electron transporting protein located in inner mitochrondrial membrane
    • Upon apoptotic stimulation, CYCS can be released from mitochondria into cytoplasm, resulting in caspase-3 activation and apoptosis
    • Measuring CYCS release from mitochondria useful for detection of onset of apoptosis in cells
      • Can also leave cells and be detectable in extra-cellular medium of apoptotic cells and serum of cancer patients
      • Level of serum CYCS may serve as prognostic marker during cancer therapy

Process:

  • Antibody specific for CYCS pre-coated onto microwells
  • CYCS protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for CYCS is added to detect captured CYCS protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
