- Fetuin A (alpha2-HS glycoprotein, AHSG) is a member of cystatin superfamily of protease inhibitors
- Highly expressed glycoprotein in various fetal tissues, mainly expressed by liver in adults
- Fetuin A is an extracellular inhibitor of transforming growth factor β is a profibrogenic stimulus in liver disease
- May be beneficial serum biomarker in detection of liver and vascular fibrosis progression in patients with non-alcoholic fatty liver disease
- Involved in functions like endocytosis, brain development and formation of bone tissue
- Fetuin A exerts effects on cardiovascular system by inhibiting insulin signaling and inducing insulin resistance
- Also contributes to onset of atherosclerosis and inhibits calcium deposition and protects from vascular calcification
Process:
- Antibody specific for Fetuin A pre-coated onto microwells
- Fetuin A protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for Fetuin A is added to detect the captured Fetuin A protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
