Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous G-CSF

  • Recognizes human G-CSF
  • Granulocyte colony-stimulating factor (G-CSF, CSF3) is a protective cytokine with anti-inflammatory effects
    • Promotes survival of granulocytic lineage cells and proliferation and migration of neutrophils and trophoblast cells
    • Acts by binding to its receptor (G-CSFR) after binding with G-CSF activates canonical Janus kinase/signal transducer, activator of transcription and Ras/Raf/MAP kinase pathways
    • Potently stimulates proliferation and release of peripheral blood progenitor cells into bloodstream and is used to treat neutropenia after chemotherapy
    • Elevated levels upon intensive exercise lead to increased neutrophil counts

Process:

  • Antibody specific for G-CSF pre-coated onto microwells
  • G-CSF protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for G-CSF is added to detect captured G-CSF protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correctional wavelength set at 630 nm
