Description
- Recognizes human granzyme B
- Granzyme B (GZMB, CG1, CSPB, CTLA1, GRB) belongs to granzyme subfamily
- Enzyme necessary for target cell lysis in cell-mediated immune responses
- Cytotoxic lymphocyte protease GZMB can promote apoptosis through direct processing and activation of members of caspase family
- Functions as processing protease of cytokines (interleukin, IL-18, IL-1a) and extracellular matrix proteins (fibronectin) and proteoglycans
- Plasma granzyme B concentrates significantly higher in patients with AD and psoriasis
- Granzyme B may play role in induction of intractable itch and dermatitis in AD
Process:
- Anitbody specific for granzyme B pre-coated onto microwells
- Granzyme B protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for granzyme B is added to detect captured granzyme B protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
