- Recognizes human GRP
- GRP (Gastrin-releasing peptide) is normally formed by mucosal cells in gastric antrum and by D cells of pancreatic islets, main function is to stimulate secretion of HCl by gastric mucosa
- HCl inhibits gastrin formation
- 148 amino acid preproprotein, following cleavage of signal peptide is further processed to produce either 27 amino acid gastrin-releasing peptide or 10 amino acid neuromedin C
- Smaller peptides regulate numerous functions of gastrointestinal and CNS (gastrointenstinal hormones, smooth muscle cell contraction, epithelial cell proliferation)
- Peptides likely to play role in human cancers (lung, colon, stomach, pancreas, breast, prostate)
Process:
- Antibody specific for GRP pre-coated onto microwells
- GRP protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for GRP is added to detect the captured GRP protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
