Invitrogen™ Silencer™ Negative Control No. 1 siRNA
Description
Ambion™ Silencer Negative Control No. 1 siRNA, In Vivo Ready has no significant sequence similarity to mouse, rat, or human gene sequences. The control has also been tested in cell-based screens and proven to have no significant effect on cell proliferation, viability, or morphology. It is highly purified as required for animal use. 250 nmol is provided.
- Nontargeting with limited sequence similarity to known genes
- Validated for use in human, mouse, and rat cells
- Functionally proven to have minimal effects on cell proliferation and viability
- HPLC purified, duplexed, and ready to use
Negative control siRNAs are essential for determining transfection efficiency and to control for the effects of siRNA delivery. In siRNA screening applications, negative control siRNAs are also critical for setting the 'hit' threshold that determines whether an siRNA is considered to have a positive, negative, or neutral effect in a particular assay. Silencer™ Negative Control No. 1 siRNA has no significant sequence similarity to mouse, rat, or human gene sequences. The control has also been tested in cell-based screens and proven to have no significant effect on cell proliferation, viability, or morphology.
Quality Control:
Silencer™ siRNAs are manufactured in a state-of-the-art facility under the highest quality standards. After HPLC purification and annealing, each siRNA is further purified utilizing a process that removes excess salt via a semipermeable membrane. The result is highly pure siRNA with minimal salt content, suitable for in vivo applications. In Vivo Ready siRNAs are then sterile filtered and tested for the presence of endotoxin. At concentrations of 50 μM in deionized water, In Vivo Ready siRNAs contain less than 0.6 mM Na+, less than 2.0 mM K+, and less than 0.1 mM Mg2+.
Accessory Product: Silencer™ siRNA Screening Control Panel (Cat. No. AM4640)
The Silencer™ siRNA Screening Control Panel includes seven negative control siRNAs, and is for those researchers performing siRNA screens that employ hundreds or thousands of siRNAs per experiment. Because negative control siRNAs may have unintended effects in a particular assay, a negative control siRNA should be carefully chosen. The Silencer™ siRNA Screening Control Panel simplifies this process. As an added benefit, a positive control siRNA to human, mouse, and rat KIF11 (Eg5) is included in the panel. Knockdown of KIF11, which encodes a kinesin family motor protein, leads to mitotic arrest. Silencer™ control siRNAs are unmodified 21-mers; they are designed to be used with Silencer™ siRNAs, BLOCK-iT™ siRNAs, and other unmodified 21mer siRNAs. If you are using other siRNA products, such as Ambion™ Silencer™ Select siRNAs or STEALTH RNAi™ siRNAs, we recommend that you use control siRNAs designed for use with those siRNAs for more reliable experimental results.
