Description
- Strep-Tactin™ TACS Agarose columns contain agarose macrobeads coated with the streptavidin variant Strep-Tactin™.
- A special crosslinking permits the use for cell isolation and depletion approaches.
- They were developed for column-based Traceless Affinty Cell Selection (Fab-TACS™/Nano-TACS™).
- In this approach, Fab fragments fused to a Twin-Strep-tag™ (Fab-Streps) or nanobodies (Nano-Streps) are bound to Strep-Tactin™ on the agarose matrix.
- After specifically capturing cells, Fab-/Nano-Streps are released from the Strep-Tactin™ Agarose by biotin addition.
- This way the cells are eluted ready for further downstream applications.
- The same principle is applicable for exosome isolation (Fab-TACS™ for exosomes).
- Alternatively to Fab-Streps/Nano-Streps, it is possible to load columns with Twin-Strep-tagged, desthiobiotinylated or biotinylated antibodies (irreversible binding).
- This makes Strep-Tactin™ TACS Agarose columns also suitable for negative cell selection or depletion approaches.
